Biotin functionalized polyethylene glycol (PEG-Biotin) is an avidin or streptavidin binding PEG derivative that is used to biotinylate biomolecules or other material surfaces. Biotin can be easily detected by biotin/streptavidin binding assay and is widely used for molecule target detection.

Desthiobiotin PEG amine (DSB-PEG-NH2) is one of Nanocs’ desthiobiotin PEG crosslinkers that have a reactive primary amine group on one PEG terminus and a desthiobiotin tag on the other PEG end. Desthiobiotin is a modified version of biotin. Compared to biotin, desthiobiotin binds to streptavidin, avidin and other biotin-binding proteins with lower affinity and it can be eluted from binding complex with much more gental conditions (i.e., by competitive displacement with regular, free biotin). As such, desthiobiotin reagent is a useful alternative to Biotin for avidin-biotin techniques in which nondenaturing elution of the labeled proteins is desired. Amine functionalized desthiobiotin can react with a number of amine reactive groups such as succinimidyl ester, carboxylic acid, aldehyde, etc. Reaction between amine and other groups allows quick and efficient conjugation of desthiobiotin tag to proteins, antibodies and other biomolecules. PEG linker between desthiobiotin and amine group offers better water solubility, reduced steric hindrance and enhanced reactivity.
