肽保护的惰性侧链氨基酸Fmoc-Val-Whaig resin树脂

根据合成肽时侧链的活性,可把自然界中 20 种氨基酸分为惰性侧链氨基酸(如:GlyLeuIleAlaVal Phe 等)和活泼侧链氨基酸(如:侧链羧基、侧链氨基、侧链硫醇基和侧链杂环基等)。前一类只需保护 α-氨基,后一种氨基酸还需保护侧链活泼基团。由于主链保护基在接肽时脱去,侧链保护基在接肽后脱去,因此这两种保护基必需具有不同的脱去条件。

【应用】

Fmoc-甘氨酸为Fmoc保护的甘氨酸,主要用于合成多肽,氨基酸保护基的不同对多肽合成的效率和产率有很大的影响。如Fmoc-甘氨酸可以用于制备一种甘氨酸衍生物缓蚀剂,通过酰胺化反应在甘氨酸分子上接入Fmoc-保护基团和疏水长链,通过杂原子(NO等或不饱和键)和含π电子的双苯环吸附于金属,疏水长链阻挡水分 子,使其在钢铁表面形成一道保护膜,达到防腐的目的。

 

【合成】

甘氨酸是简单的氨基酸,在固相合成中,要把氨基保护起来才能接肽。用 Fmoc-Cl 10%碳酸钠溶液和二氧六环条件下,低温与甘氨酸反应,可制得Fmoc-甘氨酸。实验过程中用薄层色谱监控反应进度,T.L.C 展开剂为:V(Et OH)V(CHCl3)13,试验方程式如下:

Fmoc-甘氨酸合成具体步骤如下:

3.0g 甘氨酸溶解于115ml 10Na2CO3中,冷却到-40℃11.2g Fmoc-Cl 溶解于 35.0ml 丙酮滴加到体系中,滴加完毕,冰浴搅拌30min,然后室温搅拌2.0h

用薄层色谱监控反应进度,反应完毕,倒入400ml水中,乙醚萃取两次,冷却并用浓盐酸调节 pH2 左右,析出大量白色固体,

50 ml乙酸乙酯萃取三次,无水硫酸镁干燥,减压蒸馏除去溶剂,用石油醚析出,得到 11.5g Fmoc-甘氨酸。提纯后,高效液相色谱测得产品纯度为97.3
