Description:

Maleimide functionalized polyethylene glycol, Maleimide PEG is a sulfhydryl (thiol/SH) group reactive PEG derivative that can be used to selectively modify protein, peptide or any other surfaces with availablesulfhydryl groups. The reaction between maleimide and thiol proceeds readily at pH 6.5~7.5 to form stable thioether bonds.

Physical Properties:

  • Off-white/white solid or viscous liquid depends on molecule weight;
  • Soluble in regular aqeous solution as well as most organic solvents;

Storage Conditions:

  • Store at -20 0C, dessiccated. Avoid frequent thaw and freeze.

Reaction Procedures:

Generally, a 10- to 20-fold molar excess of Maleimide PEG over the amount of thiol-containing material results in sufficient conjugation.

Materials Required:

  • Pegylation buffer, PBS buffer, pH 7 or other thiol-free buffer at pH 7.
  • Maleimide stock solution: 100 mg in 1 mL conjugation buffer.
  • Washing solution: Distilled water or any aqueous buffer.

Reaction Steps:

Dissolve targeted materials in conjugation buffer. Estimate the concentration thiol groups on the targeted materials. Add maleimide PEG stock solution to the targeted conjugation materials with the final concentration keep at least 10 mg/mL. 10~50 molar excess of PEG maleimide needed for optimal conjugation; Allow mixture agitates at room temperature for 2~4 hours at room temperature or overnight at 4 0C.Conjugate can be purified either by size exclusion chromatography or dialysis.

A.以人 NA19240 基因组 DNA（Coriell Institute）为起始样本，使用 NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒和 Illumina PCR-free 文库制备试剂盒分别制备 PCR-free 文库，350 bp 插入片段作为筛选标准。
B.分别使用基于酶法片段化的 NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒和 Roche Sequencing Kapa HyperPlus 文库制备试剂盒，制备 150 – 200 bp 插入片段的 PCR-free 的文库，不进行片段筛选。根据生产商推荐方法，NEBNext 和 Kapa 试剂盒分别使用了 NEBNext 多样本接头引物试剂盒 1（Unique 双端 Index 引物，UMI 接头，适用于 DNA）和 Kapa Dual-Indexed Adaptors。

图2：使用 NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒，实现人类基因组均一覆盖。相较于 Illumina DNA PCR-free 文库制备试剂盒，NEBNext Ultra II FS DNA PCR-free 文库制备试剂盒可实现更高的基因组覆盖度。根据生产商推荐方法，以 250 ng NA19240 基因组 DNA 为起始样本，分别由 2 位实验员制备文库，使用 NovaSeq 6000 S4 v1.5 flow cell（PE 2 x 150 bp）测序。Reads 去接头（fastp 0.20.0）后比对至端粒到端粒 chm13 参考基因组（draft 1，bwa-mem 0.7.17），并标记重复（samblaster 0.1.24）。从每个 bam 中随机抽取 2.8 B Reads（sambamba view -s 0.6.8）。使用 mosdepth（v0.3.1，-F 772）评估过滤水平和初级覆盖度，并绘制常染色体或线粒体基因组图。图中标示出了预期覆盖度（num_reads*read length/genome size）。两位实验员在使用 Illumina PCR-free 文库制备试剂盒时覆盖度存在显著差异，而使用 FS DNA PCR-free 文库制备试剂盒时不存在该问题，结果非常一致。

Description

DSPE-PEG-COOH is one of Nanocs reactive phospholipid PEG conjugates that can react with primary amine groups. DSPE (1,2-distearoyl-sn-glycero-3-phosphoethanolamine) is an 18 carbon phospholipid that is highly hydrophobic. PEG linker, on the other hand, offers good hydrophilicity and water solubility. Carboxylic group (-COOH) on the other PEG terminal reacts with primary amine groups using EDC chemistry. PEG DSPE thus can be easily incorporated into liposome and other nanoparticles via the reaction of carboxylic acid and amine. DSPE PEG COOH is a commonly used reactive phospholipid to conjugate antibodies, proteins, peptides or other substrates to the surface of liposome and other lipid PEG nanoparticles. Pegylated phospholipids have longer blood circulation time and higher stability for encapsulated molecules. Nanocs has developed a comprehensive collection of chemical reactive phospholipid PEG products that have high purity, various molecular weights and excellent chemical reactivity. Description:

Polystyrene Particles from Nanocs have narrow size distribution yet with multiple size selection from 20 nm to 10 microns. These particles are suspended in 1% aqeuous solution. Standard deviation of these particles is in the range of 10% of listed size parameter. These particles have wide applications in diagnostic reagents and bioassay development. They can be further modified with different surface coatings and functional groups. Nanocs surface modified polystyrene particles provide more choices for in-vitro or in-vivo applications of those nanoparticles. Customer conjugation and modification is available upon request.
