Fetal Bovine Serum优级plus胎牛血清来源于无菌采集的健康胎牛血清，适用于大部分原代细胞和部分免疫细胞，以及部分难贴壁细胞，典型代表有：UVEC、HUVEC；THP-1、REC-1、Raw264.7；293T、MRC-5。Bestop™胎牛血清在GMP净化车间全自动灌装生产，经过三级0.1μm过滤，在严格质量控制体系下，经过病毒、支原体、内毒素等多项测试，无细菌、真菌、病毒、支原体及黑胶虫等污染，内毒素含量≤ 10EU/ml，血红蛋白含量≤ 200mg/L，批量大、批间差小，具有高质量和高稳定性。

使用方法：

1) Fetal Bovine Serum优级plus胎牛血清从低温冰箱取出后，于 2-8℃冰箱放置 12-24h， 使之部分融解，而后置于室温下使之全部融解。在融解过程中需及时规则地轻柔混匀。血清在溶解过程中可能会析出絮状物，这些成分主要是血清中的纤维蛋白，是正常现象，对后续的培养无害，不需去除。

2) 基础培养基中胎牛血清的使用浓度一般为 5-20%，请根据具体细胞种类及实验要求按需添加。

3) 血清分装建议在无菌环境下进行。

注意事项：

1) 本产品避免反复冻融。 

2) 本产品若放于 2℃~8℃冷藏条件下，保存时间建议不超过 1 个月。 

3) 本产品未经热灭活，需根据实验需要判断是否进行热灭活。热灭活时请将血清置于56℃水浴30分钟。需要注意，在热灭活过程中，会导致血清中某些成分被破坏。 

4) 本产品在使用过程中请穿实验服，佩戴一次性手套，并注意无菌操作，防止细菌污染。

Additionally, it supports growth of chicken embryo fibroblasts under serum-free conditions. Virus production can then be conducted under serum-free conditions without the addition of albumin or other supplements.


The Diploid SRM system is composed of two different kits:

1. Diploid Growth Serum-Reduced Medium (SRM) kit: A basal medium and 100X growth supplement designed for cell expansion

2. Diploid Production Serum-Free Medium (SFM) kit (this page): A basal medium and 100X production supplement designed for virus production


Human cells have traditionally been used to manufacture a variety of vaccines, such as the varicella zoster, polio, smallpox, hepatitis A, measles, mumps, and rubella vaccines. Traditionally, the only way to culture these cells has been to use classical media with 10% serum for growth and 1–5% human albumin (human sourced or recombinant) for virus production. These are two of the highest cost components in vaccine production. The Diploid SRM system helps reduce or eliminate both components while maintaining high virus titers.


Cost savings:

Reduction or elimination of serum can significantly reduce cost for a vaccine manufacturing process. Premium FBS is expensive with high price volatility. Given the typical cost pressures on vaccine production, both economic drivers make vaccine manufacturing to a target price extremely challenging. Diploid Growth SRM reduces the impact of this volatility in price and offers a level of cost consistency not available with processes that use serum in high volumes.


In addition, serum requires frozen storage. A 10% serum concentration in growth media can result in high frozen storage requirements for a vaccine process. A process requiring 1,000 L of growth medium would require the frozen storage of 100 L of serum. The Diploid Growth SRM kit comes with a 100X frozen supplement, enabling an 80% reduction in frozen storage requirements. 1,000 L of Diploid Growth SRM would only need 20 L of frozen storage: 10 L for FBS and 10 L for the 100X supplement.


Albumin is commonly added to virus production media. With the Diploid Production SFM, viruses can be produced at comparable titers without any additional supplementation.

UElandy无血清（Serum Free）细胞冻存液为即用型产品，不含血清和动物来源性蛋白。因此，不仅降低了各类细菌、病毒、支原体等污染风险，保证冻存细胞的安全，而且减少外源性蛋白对细胞正常生长和分化的影响。此外，无需程序降温，可长期-80℃保存细胞，适用于绝大多数哺乳动物细胞的冻存。

注意事项

1. 使用前确保细胞冻存液彻底融化，并轻轻混匀。融化后的细胞冻存液置于4ºC保存，三个月内有效。
2. 本产品含有酚红指示剂，长时间敞口会导致PH变化，并出现沉淀。故使用过程中请尽量缩短敞口时间，使用完毕后盖紧盖子保存，如有必要可适当分装使用。若使用一段时间后出现少量沉淀，经测试对细胞冻存无影响，去除沉淀可采取离心取上清、0.22 μM孔径过滤器过滤等方法去除。
3. 细胞冻存前请确保其状态良好，活细胞比例宜大于90%。
4. 本产品含DMSO，部分对DMSO敏感的细胞，建议进行预实验。
5. 对于某些珍贵的细胞，建议同时使用含FBS的常规冻存液进行对比测试，确认性能后再进行正式冻存。
6. 冻存细胞分装后应尽快移入-80ºC超低温冰箱内，减少在外存放时间。

注意事项

1. 血清解冻后应尽快用完，尽量避免反复冻融；如果不能短期内使用完毕，解冻后请适当分装。血清结冰时体积会增加约10%，因此在分装血清时须使分装瓶预留一定体积空间，否则易导致分装瓶冻裂而发生污染。
2. 血清解冻时应将血清从-15~-40℃的低温冰箱中取出放入4℃冰箱解冻1天，待全部解冻后再分装。解冻过程中请不时摇匀（小心勿造成气泡），使血清成分和温度均匀，从而减少沉淀的产生
3. 切勿直接将血清从-15℃或更低温度进入37℃水浴解冻，这样因温度改变太大，容易造成蛋白质凝集而出现沉淀，从而使用血清的质量下降。
4. 血清中的絮状沉淀物主要是解冻后血清中纤维蛋白及4℃长时间保存后血清中的脂蛋白变性造成的，不会影响血清本身的质量，可不用处理，也可400×g离心5min去除。

常见问题解答：

◆这款胎牛血清的试用范围？

适用于一般细胞，肿瘤细胞，部分干细胞等。

◆如何储存和解冻血清才不会使产品质量受损?
血清解冻时应将血清从-15~-70℃的低温冰箱中取出放入4℃冰箱解冻1天，待全部解冻后再分装。解冻过程中请不时摇匀（小心勿造成气泡），使血清成分和温度均匀，从而减少沉淀的产生。若一次无法用完一瓶，建议无菌分装血清至恰当的灭菌容器内，再放回冷冻，并避免反复冻融。需要长期保存的血清必须储存于-20℃ – 70℃ 低温冰箱中。4℃冰箱中保存时间切勿超过1个月。切勿将血清在 37℃放置太久,否则血清会变得浑浊,同时血清中的有效成分会被破坏，而影响血清质量。

◆有时在血清中见到的絮状沉淀可能是些什么物质？
主要是血清中的脂蛋白变性及解冻后血清中纤维蛋白造成,这些絮状物不会影响血清本身的质量，可用400 xg离心5 min去除，也可不用处理。

◆如何避免血清中产生沉淀?
(1)解冻血清时，请随时将之摇晃均匀，使温度及成分均一，减少沉淀的发生。
(2) 血清分装冻存时，须规则摇晃均匀(小心勿造成气泡)，使温度与成分均一。
(3) 勿直接由-20℃直接至37℃解冻，因温度改变太大，容易造成蛋白质凝结而发生沉淀。
(4) 勿将血清置于37℃太久，否则血清会变得浑浊，同时血清中许多较不稳定的成份也会因此受到破坏，从而影响血清的品质。
(5) 血清的热灭活非常容易造成沉淀物的增多，若非必要，可以无须做此步骤。
(6) 若必须做血清的热灭活，请遵守56℃，30分钟的原则，并且随时摇晃均匀。温度过高，时间过久或摇晃不均匀，都会造成沉淀物的增多。

◆在使用前是否需要对血清进行过滤？
一般厂商提供的血清为无菌,无需再过滤除菌。如发现血清有悬浮物,则可将血清加入培养液内一起过滤,切勿直接过滤血清。

◆自然溶化好的胎牛血清放在室温下一晚还能用吗？
室温不可以，建议4℃保存。

◆加到培养基中的血清必须灭活吗？
不是必须的，看做什么实验。如果用于培养普通细胞，血清一般是不需要灭活的，这样可以有效保存血清中的生长因子。但是如果用于培养一些表面具有补体受体的细胞，如内皮细胞，血清是需要灭活，一般是56℃，30min。

◆热灭活对血清的作用是什么？
热灭活步骤能够灭活补体系统。补体会参与以下事件中：溶解细胞事件，平滑肌收缩，肥大细胞与血小板释放组胺，吞噬作用增强，趋化作用，淋巴细胞与巨噬细胞的活化等。

◆细胞培养 中出现黑点是污染吗?如何处理?
一旦您在细胞培养的过程中发现有黑点生成，首先，要肉眼观察培养基是否混浊，然后，在镜下观察培养细胞生长的状态，黑点是否游动。如果细胞被污染，微生物则会大量繁殖，培养基就会迅速变黄、变混浊。污染包括细菌污染、支原体污染等。
如果在镜下观察细胞，生长状态良好，与黑点出现前相比，没有任何变化，那么，黑点的出现可能与以下几种情况有关：
(1)细胞生长过老，破碎的细胞残骸;
(2)血清质量不好，反复冻融的结果;
(3)配制培养基的pH值偏高，不宜细胞生长;
(4)配制培养基的水质、容器不合格。可应用离心、过滤的方法去除这些黑点;
(5)培养原代细胞中出现小黑点，可能是原代组织中的杂质，多次传代可以消除。

◆细胞培养中如何防止黑点生成?
(1) 尽可能地减少血清冻融次数。
(2) 培养基无需37℃水浴。
(3) 培养基保持最佳PH值7.0- 7.2。
(4) 严格控制配制培养基的水质，容器定期刷洗。
(5) 掌握细胞传代的最佳时机，细胞切勿生长过老。

Principle
