鬼笔环肽是从致命的伞形毒蕈蘑菇中分离出来的一种毒素。它是特异性结合于F-肌动蛋白的双环肽。因此用荧光染料标记的鬼笔环肽可以非常方便的研究F-肌动蛋白的分布。鬼笔环肽内部,在半胱氨酸和色氨酸之间含有不常见的硫醚桥形成内环结构。在pH升高时,该硫醚被裂解,鬼笔环肽失去对肌动蛋白的亲和力。
YF®染料是我司开发的一系列新一代荧光染料,与其他荧光染料相比,在亮度,光稳定性和水溶性方面具有综合优势。荧光标记的鬼笔环肽可在纳摩尔水平染色F-肌动蛋白。在各种植物细胞或动物细胞中,标记的鬼笔环肽对大、小细丝具有相似的亲和力,平均每个肌动蛋白亚基结合一个鬼笔环肽分子。不同于抗体,鬼笔环肽与肌动蛋白的结合亲和力在不同物种间没有显着变化。非特异性染色可以忽略不计,染色和未染色区域之间的对比度非常大。鬼笔环肽将单体/聚合物的平衡转向聚合状态,将聚合临界浓度降低至30倍。鬼笔毒肽类(Phallotoxins)可通过抑制细胞松弛素的解聚,碘化钾和升高的温度,稳定F-肌动蛋白。因为鬼笔环肽缀合物很小,大约直径12-15埃,分子量< 2000 Dal,多种肌动蛋白结合蛋白,包括肌球蛋白,原肌球蛋白和后肌钙蛋白依然可以和鬼笔环肽标记的肌动蛋白结合。更重要的是,鬼笔环肽标记的肌动蛋白丝保持功能,标记甘油肌纤维仍然收缩,标记的肌动蛋白丝仍然可以继续移动。而且荧光标记的鬼笔环肽也可用于对细胞中F-肌动蛋白进行定量研究。

为什么我到的是空管?

本产品为冻干粉形式,微量不易观察,使用前请瞬时离心,加适当溶剂溶解后使用。

鬼笔环肽染色细胞骨架过程中,能使用甲醇固定细胞吗?

甲醇会破坏Actin蛋白,因此不能使用含有甲醇的固定液,并且使用不含甲醇的甲醛。

为什么用鬼笔环肽结合F-actin标记石蜡切片,没有看到信号?
当细胞和组织经由二甲苯或丙酮之类的溶剂处理时(例如组织切片脱石蜡期间),它通过阻止鬼笔环肽结合的方式来影响F-actin。可以使用通常不用有机溶剂洗涤的冷冻切片代替石蜡切片,或者可以使用抗肌动蛋白抗体。

鬼笔环肽染料能重复利用吗?
正常情况下染色液在使用后,可能会影响染色效果,最好不要重复使用;我们的鬼笔环肽按说明书添加,浓度较低,这是染料不能重复利用的原因。

鬼笔环肽染色F-Actin, 有的染上,有的没染上,并且染色信号较弱怎么办?
(1)可能是染料浓度低或孵育时间不足,建议增加染料浓度或培养时间。
(2)可能是在错误的波长下分析细胞,建议重新检查过滤器设置是否正确。

不进行透化步骤,鬼笔环肽直接染色固定后的细胞可以吗?
荧光标记的phalloidins,只能用于在固定透化后的组织切片、细胞培养和无细胞实验中染色F-actin。

染色后,没有荧光信号怎么办?
(1)检查储液、工作液配制是否有误。
(2)固定及透化步骤尤为重要,使用新鲜配置的4%多聚甲醛进行固定,使用新鲜配置的0.5% Triton X-100 进行透化。摸索最佳的固定及透化时间。
(3)摸索最佳染色工作液浓度及染色时间。

可以用于做流式细胞吗?
该产品主要是用于微丝的染色,一般用荧光显微镜观察结果,不推荐用流式检测。

可以用普通荧光显微镜观察吗?
普通的荧光显微镜也是可以检测的,激光共聚焦拍照效果会更好一些。

探针的浓度是多少?
请按照说明书的稀释比例进行使用,具体浓度信息未知。

Rhodamine 123 是一种可透过细胞膜的阳离子绿色荧光染料,可作为线粒体膜电位的指示剂,其在正常细胞中能够依赖线粒体膜电位进入线粒体基质,荧光强度弱。而在凋亡发生时,线粒体膜完整性破坏,线粒体膜通透性转运孔开放,引起线粒体膜电位的降低,Rhodamine 123 重新释放出线粒体,从而发出强黄绿色荧光,广泛用作检测线粒体膜电位的荧光探针,并且对细胞没有任何毒性。可用荧光显微镜、荧光光度计或流式细胞仪检测,通过荧光信号的强弱来检测线粒体膜电位的变化和细胞凋亡的发生,可用于培养的细胞或从组织中提取出的线粒体的膜电位的检测。

Description:

DBCO (Dibenzocyclooctyne) is a cycloalkyne that can reacts with azides via strain-promoted 1,3-dipolar cycloaddition in aqueous solution, a bioorthogonal reaction also termed Cu-free click reaction.  This reaction has excellent selectivity and biocompatibility, such that the complimentary reagents can form covalent bonds with richly functionalized biological systems. The Cu-free click reaction has been a powerful tool in catalyst-free bioconjugation. DBCO reagents posses fast kinetcs and stability in aqueous buffer and they can be used to label azide-modified biomolecules with high specificity and reactivity.

Copper-free Click Reaction Procedures:

Prepare the azide-containing sample in reaction buffer. Add DBCO conjugate to azide containing sample. Incubate the reaction at room temperature for 2-4 hours or 2-12 hours at 4 degree celcius. The reaction is now ready for purification.
