产品介绍
鬼笔环肽是从致命的伞形毒蕈蘑菇中分离出来的一种毒素。它是特异性结合于F-肌动蛋白的双环肽。因此用荧光染料标记的鬼笔环肽可以非常方便的研究F-肌动蛋白的分布。鬼笔环肽内部，在半胱氨酸和色氨酸之间含有不常见的硫醚桥形成内环结构。在pH升高时，该硫醚被裂解，鬼笔环肽失去对肌动蛋白的亲和力。
YF®染料是我司开发的一系列新一代荧光染料，与其他荧光染料相比，在亮度，光稳定性和水溶性方面具有综合优势。荧光标记的鬼笔环肽可在纳摩尔水平染色F-肌动蛋白。在各种植物细胞或动物细胞中，标记的鬼笔环肽对大、小细丝具有相似的亲和力，平均每个肌动蛋白亚基结合一个鬼笔环肽分子。不同于抗体，鬼笔环肽与肌动蛋白的结合亲和力在不同物种间没有显着变化。非特异性染色可以忽略不计，染色和未染色区域之间的对比度非常大。鬼笔环肽将单体/聚合物的平衡转向聚合状态，将聚合临界浓度降低至30倍。鬼笔毒肽类（Phallotoxins）可通过抑制细胞松弛素的解聚，碘化钾和升高的温度，稳定F-肌动蛋白。因为鬼笔环肽缀合物很小，大约直径12-15埃，分子量< 2000 Dal，多种肌动蛋白结合蛋白，包括肌球蛋白，原肌球蛋白和后肌钙蛋白依然可以和鬼笔环肽标记的肌动蛋白结合。更重要的是，鬼笔环肽标记的肌动蛋白丝保持功能，标记甘油肌纤维仍然收缩，标记的肌动蛋白丝仍然可以继续移动。而且荧光标记的鬼笔环肽也可用于对细胞中F-肌动蛋白进行定量研究。

常见问题解答：

◆为什么我拿到的是空管？

本产品为冻干粉形式，微量不易观察，使用前请瞬时离心，加适当溶剂溶解后使用。

◆鬼笔环肽染色细胞骨架过程中，能使用甲醇固定细胞吗？

甲醇会破坏Actin蛋白，因此不能使用含有甲醇的固定液，并且使用不含甲醇的甲醛。

◆为什么用鬼笔环肽结合F-actin标记石蜡切片，没有看到信号？
当细胞和组织经由二甲苯或丙酮之类的溶剂处理时（例如组织切片脱石蜡期间），它通过阻止鬼笔环肽结合的方式来影响F-actin。可以使用通常不用有机溶剂洗涤的冷冻切片代替石蜡切片，或者可以使用抗肌动蛋白抗体。

◆鬼笔环肽染料能重复利用吗？
正常情况下染色液在使用后，可能会影响染色效果，最好不要重复使用；我们的鬼笔环肽按说明书添加，浓度较低，这是染料不能重复利用的原因。

◆鬼笔环肽染色F-Actin, 有的染上，有的没染上，并且染色信号较弱怎么办？
（1）可能是染料浓度低或孵育时间不足，建议增加染料浓度或培养时间。
（2）可能是在错误的波长下分析细胞，建议重新检查过滤器设置是否正确。

◆不进行透化步骤，鬼笔环肽直接染色固定后的细胞可以吗？
荧光标记的phalloidins，只能用于在固定透化后的组织切片、细胞培养和无细胞实验中染色F-actin。

◆染色后，没有荧光信号怎么办？
（1）检查储液、工作液配制是否有误。
（2）固定及透化步骤尤为重要，使用新鲜配置的4%多聚甲醛进行固定，使用新鲜配置的0.5% Triton X-100 进行透化。摸索最佳的固定及透化时间。
（3）摸索最佳染色工作液浓度及染色时间。

◆可以用于做流式细胞吗？
该产品主要是用于微丝的染色，一般用荧光显微镜观察结果，不推荐用流式检测。

◆可以用普通荧光显微镜观察吗？
普通的荧光显微镜也是可以检测的，激光共聚焦拍照效果会更好一些。

◆探针的浓度是多少？
请按照说明书的稀释比例进行使用，具体浓度信息未知。

产品介绍
Rhodamine 123 是一种可透过细胞膜的阳离子绿色荧光染料，可作为线粒体膜电位的指示剂，其在正常细胞中能够依赖线粒体膜电位进入线粒体基质，荧光强度弱。而在凋亡发生时，线粒体膜完整性破坏，线粒体膜通透性转运孔开放，引起线粒体膜电位的降低，Rhodamine 123 重新释放出线粒体，从而发出强黄绿色荧光，广泛用作检测线粒体膜电位的荧光探针，并且对细胞没有任何毒性。可用荧光显微镜、荧光光度计或流式细胞仪检测，通过荧光信号的强弱来检测线粒体膜电位的变化和细胞凋亡的发生，可用于培养的细胞或从组织中提取出的线粒体的膜电位的检测。

Description:

DBCO (Dibenzocyclooctyne) is a cycloalkyne that can reacts with azides via strain-promoted 1,3-dipolar cycloaddition in aqueous solution, a bioorthogonal reaction also termed Cu-free click reaction.  This reaction has excellent selectivity and biocompatibility, such that the complimentary reagents can form covalent bonds with richly functionalized biological systems. The Cu-free click reaction has been a powerful tool in catalyst-free bioconjugation. DBCO reagents posses fast kinetcs and stability in aqueous buffer and they can be used to label azide-modified biomolecules with high specificity and reactivity.

Copper-free Click Reaction Procedures:

Prepare the azide-containing sample in reaction buffer. Add DBCO conjugate to azide containing sample. Recommendation: Add 1 mole equivalent of limiting reagent to 1.5-3.0 mole equivalents of highest abundance reagent. Incubate the reaction at room temperature for 2-4 hours or 2-12 hours at 4 degree celcius. The reaction is now ready for purification.

Dextrans are naturally synthesized polysaccharides with good water solubility, low toxicity and varied molecular weights. Rhodamine B labeled Dextran, also called TRITC or RB dextran, is a pink colored dextran conjugate that emits red  fluorescent light upon excited by light at 540-550 nm. Rhodamine B Dextran conjugates from Nanocs have high fluorescent intensity, essentially free of non-conjugated dyes. These products have been successfully used for imaging lymph nodes, neurons, blood vessels and many biological processes. They are also used for long-term tracers for live cells.

Nanocs provides a variety of chemically functionalized and bioconjugated dextran products with molecular weight ranged from 3,000 Da to 2,000 kDa. These functional dextrans were purified by size chromatography to remove non-conjugated molecules and ensure adequate applications both in-vitro and in-vivo. Polystyrene Particles from Nanocs have narrow size distribution yet with multiple size selection from 20 nm to 10 microns. These particles are suspended in 1% aqeuous solution. Standard deviation of these particles is in the range of 10% of listed size parameter. These particles have wide applications in diagnostic reagents and bioassay development. They can be further modified with different surface coatings and functional groups. Nanocs surface modified polystyrene particles provide more choices for in-vitro or in-vivo applications of those nanoparticles. Customer conjugation and modification is available upon request.
