Description:

Nanocs provides a variety of chemically functionalized and bioconjugated dextran products with molecular weight ranged from 3,000 Da to 2,000 KDa. These functional dextrans were purified by size chromatography to remove non-conjugated molecules and ensure adequate applications both in-vitro and in-vivo.

Dextrans are naturally synthesized polysaccharides with good water solubility, low toxicity and varied molecular weights. Dextrans modified with dyes and other biomolecules have been successfully used as long-term tracers for live cells. Nanocs also provides customerized dextran products for your special needs.

Description:

Amino PEG Acid (NH2-PEG-COOH) is one of  Nanocs reactive heterobifunctional PEG derivatives that have an amine group on one PEG end and a carboxylic acid group on the other PEG terminus. Amino PEG acid can be used to modify proteins, peptides and other materials via amino or other acid reactive chemical groups. PEGylation can increase solubility and stability and reduce immunogenicity of peptides and proteins. It can also suppress the non-specific binding of charged molecules to the modified surfaces.

Pyrene PEG Aminooxy, Pyrene-PEG-ONH2:

Pyrene PEG Aminooxy (Pyrene-PEG-ONH2 is one of Nanocs’ carbonyl reactive fluorescent dyes that can label polysaccharides and glycolated proteins or enzymes. Pyrene is a unique fluorescent dye that has exceptionally long excite-state lifetimes (>100 nanoseconds). The long excited-state life time of this dye makes it useful for time-resolved fluorescent immonoassays, probing oxygen in cells and lipid vesicles. Aminooxy (-ONH2) functionalized pyrene PEG dye reacts readily with aldehyde or ketone groups to form a stable oxime bond. Reaction between aminooxy and carbonyl group allows easy and quick conjugation of pyrene to polysaccharides and glycoproteins through aldehyde groups introduced by periodate oxidation. PEG linker between Pyrene and NHS group offers better water solubility, less steric hindrance and enhanced stability. Compared to other common fluorescent dyes, Nanocs’ reactive fluorescent PEG dyes are brighter, more stable and easier to use. All our reactive fluorescent PEG dyes can be used directly in aqueous buffer without need adding any organic solvents.

Methoxy PEG propionaldehyde is a stable aldehyde PEG that can be used for protein, antibody, peptides or carbohydrate N-terminal amine group pegylation reagent. The reaction between aldehyde group with the α-amine at the N-terminus produces an intermediate Schiff base. Further reduction with hydride will form a stable C-N bond. PEG aldehydes react with n-terminus amine groups at a pH of from 5.0 to 9.5. Higher pH will result in multiple Pegylation with both terminal and lysine groups.

Reaction Procedures:

Generally, a 5- to 10-fold molar excess of PEG aldehyde over the amount of amine-containing material results in sufficient conjugation.

Materials Required:

  • Pegylation buffer: Amine-free aqueous buffer, pH 5.0~6.5.
  • PEG aldehyde stock solution: 50 mg in 1 mL conjugation buffer.
  • Washing solution: Distilled water or any aqueous buffer.

Reaction Steps:

Dissolve targeted materials in pegylation buffer. Estimate the concentration of primary amine groups on the targeted materials. Add PEG aldehyde stock solution to the targeted conjugation materials with the final concentration keep at least 10 mg/mL. Allow mixture agitates at room temperature for 2~4 hrs at room temperature or overnight 4 0C. After reaction, C=N bond can be reduced by sodium borohydride. Conjugate can be purified either by size exclusion chromatography or dialysis.

These gold nanoparticles have spherical shape, completely soluble in water, have been used widely for the development of diagnostic probes, therapeutic agents and drug delivery vehicles. The optical and electronic properties of gold nanoparticles are tunable by changing the size, shape, surface chemistry and aggregate state.
