Gibco™ Human Mammary Epithelial Cell Basal Medium (formerly ''''Medium 171''人乳腺上皮细胞基础培养基)

This medium contains essential and non-essential amino acids, vitamins, other organic compounds, trace minerals, and inorganic salts. It does not contain antibiotics, antimycotics, hormones, growth factors, or proteins. It is HEPES and bicarbonate buffered and designed for use in an incubator with an atmosphere of 5% CO2/95% air.

To support plating and long-term proliferation of normal human mammary epithelial cells, Human Mammary Epithelial Cell Basal Medium must be supplemented with Mammary Epithelial Growth Supplement (MEGS) (Cat. No. S0155). Each 5-mL bottle of MEGS contains all of the growth factors, hormones, and tissue extracts necessary for the culture of normal human mammary epithelial cells and is the correct amount of supplement for a 500 mL bottle of Human Mammary Epithelial Cell Basal Medium.

When a 500 mL bottle of medium is supplemented with MEGS, the final concentrations of the MEGS components in the supplemented medium are: BPE, 0.4% v/v; recombinant human insulin-like growth factor-I, 0.01 μg/mL; hydrocortisone, 0.5 μg/mL; and recombinant human epidermal growth factor, 3 ng/mL.

Rat Mammary Epithelial Cells are characterized by immunofluorescent staining with antibodies of E-cadherin and ZO-1. Rat Mammary Epithelial Cells are negative for bacteria, yeast, fungi, and mycoplasma. Cells can be expanded for 3-7 passages at a split ratio of 1:2 or 1:3 under the cell culture conditions specified by AcceGen. Repeated freezing and thawing of cells is not recommended.

Standard biochemical procedures performed with epithelial cell cultures include the assays of cell to cell adhesion and migration, RT-PCR, Western blotting, immunoprecipitation, immunofluorescent staining or immunofluorescent flow cytometry or generating cell derivatives for desired research applications.；

使用说明：
◉使用前准备
（1）15mL无菌离心管、移液器、移液管、无菌枪头等表面消毒后放入超净工作台中紫外照射30min。
（2）提前30min从4℃冰箱取出*培养基平衡至室温，提前从-20℃冰箱取出所需体积的细胞外基质胶冰上融化。
◉乳腺原代细胞培养
（1）采用预冷的DMEM/F12培养基将细胞外基质胶（Corning#356231）按1∶ 50比例稀释，至少提前30min 将稀释后的基质胶预铺在培养瓶/板底部表面，并使其*覆盖底部表面，30~60 min后吸干备用。
（2）将分离并计数后的细胞悬液参考表1中细胞数量接种于培养瓶/板，补加*培养基至所需体积，轻轻摇晃混匀，表面消毒后放置于培养箱，37℃、5%CO2条件下进行培养。
（3）原代细胞初次接种后2-3天内勿动（利于细胞贴壁）。

◉原代乳腺上皮细胞传代
（1）镜下观察细胞形成克隆，且汇合度达80~90%时即可传代。
（2）培养箱中取出细胞，弃去旧培养液，加入适量TrypLE*Express酶（Gibco#12605010）37℃孵育，10~20min后取出，轻拍培养瓶/板侧面，显微镜下观察细胞消化情况。
（3）细胞消化至细胞变圆并开始脱落时用含10%胎牛血清的DMEM培养基终止消化，并将细胞转移至离心管中，室温，300g，离心5min。
（4）弃上清，以1-2mL*培养基重悬细胞，活细胞计数，将细胞悬液按适合的细胞密度接种于预铺有基质胶的培养瓶/板底部，预铺方法同上。
（5） 补加*培养基至所需体积，轻轻摇晃混匀，表面消毒后置于培养箱，37℃、5%CO2条件下继续培养。

使用说明
◉使用前准备
(1) y射线照射过的NIH-3T3细胞（40Gy辐照）。
(2)15mL无菌离心管、移液器、移液管、无菌枪头等表面消毒后放入超净工作台中紫外照射30min。提前30min从4℃冰箱取出原代细胞培养基平衡至室温。
◉乳腺原代上皮细胞培养
(1)人乳腺组织经消化酶消化后，经100微米筛网过滤后计数，按照表1中的细胞数接种在合适的培养器皿中，加入y 射线辐照后的NIH-3T3细胞（添加滋养细胞数量如表1和表2所示）。
(2)原代细胞初次接种后2-3天勿动（利于细胞贴壁）），培养过程中若培养基颜色变黄但细胞未长满时可换半液，镜下观察细胞未长满但3T3细胞已不足时，适量补充y射线辐照后的3T3细胞（一般补加初始数目的一半））。

注∶以上表格仅供参考。倍增时间>48小时则定义为增殖较慢的细胞∶对干增殖较快细胞.可以综合倍增时间、接种器皿的规格以及培养周期来估算接种的细胞数量。具体实验需按具体情况对待。
◉乳腺原代上皮传代
(1)镜下观察细胞形成克隆，且汇合度达到80~90%时即可传代。
(2)培养箱中取出细胞，弃旧培养液，0.25%胰酶洗涤30秒后吸尽，再加入适量TrypLE™ Express酶 (Gibco#12605010)，37℃孵育，10-20min后拿出轻拍培养瓶/板侧面，显微镜下观察细胞消化情况。
(3)待消化至细胞变圆并开始脱落时用含10%胎牛血清的DMEM培养基终止消化，并将细胞转移至离心管中， 300g, 5min离心。
(4)弃上清，以1-2mL本细胞培养基产品重悬细胞，活细胞计数，将细胞悬液按适合的细胞密度接种于培养瓶/ 板。
(5)之后,加入丫射线辐照后的NIH-3T3细胞(不同规格培养瓶/板底面积、接种原代细胞数量、添加滋养细胞 数量如表1和表2所示)，在培养板/孔中补加乳腺原代上皮细胞培养基至所需体积，轻轻揺晃混匀，表面消毒后置培养箱，37℃、5% CO2条件培养其他步骤同上。
注意事项：

1.本产品在使用前需平衡至室温。
2.本产品中含有胎牛血清和原代细胞专用抗生素，如无特别需要不用额外再添加血清和双抗，可直接使用。
3.为保持本产品的理想使用效果，不宜将其过夜放置于室温或较高的温度环境中。
4.请于保质期内使用本产品，超出保质期，必须放弃使用。
5.原代细胞初次接种时会贴壁较慢和贴壁不牢，若无必要，尽量减少取出观察的次数，建议2-3天一次。
6.传代消化时切记不要消化过度，对细胞造成损伤，影响细胞状态。
7.接种细胞时注意细胞密度，不可过稀或过密。
8.使用产品时应注意无菌操作，避免污染。
9.本产品仅用于科研用途，不能用于体外诊断。
