Description：The human retina is a multi-layered structure lining the rear of the eye. It consists of photoreceptor cells and retinal pigment epithelial cells (RPE). RPE situated between the neurosensory retina and the choroid, form the outer blood-retina barrier that controls the chemical composition of the subretinal space. RPE has long been recognized to differ from many other epithelia in the polarity of a number of proteins and whereas almost all other epithelia face an apical lumen devoid of matrix, the RPE apical plasma membrane is in contact with the extracellular matrix. During early embryonic development, RPE spontaneously transdifferentiate into neural retinal tissue. RPE play a critical role in the development and maintenance of adjacent photoreceptors. They generate a number of reactive oxygen species when illuminated with light and their proliferation is an important step in the pathogenesis of ocular diseases such as proliferative retinopathy. HRPEpiC from AcceGen Research Laboratories are isolated from human retina. HRPEpiC are cryopreserved at passage one and delivered frozen. Each vial contains >5×10^5 cells in 1 ml volume. HRPEpiC are characterized by immunofluorescent method with antibodies to cytokeratin-18, cytokeratin-19 and fibronectin. HRPEpiC are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HRPEpiC are guaranteed to further expand for 15 population doublings in the condition provided by AcceGen Research Laboratories.；

NK-5962是光电染料视网膜假体(OUReP)的关键组成部分。


在安全性和有效性测试中，NK-5962在医疗设备生物学评价的所有测试中都是安全的(ISO 10993)，即使在黑暗条件下也能有效防止视网膜细胞死亡。


将光电染料偶联聚乙烯薄膜长期植入遗传性视网膜营养不良(RCS)大鼠视网膜下间隙，可防止邻近视网膜组织神经元的凋亡。通过玻璃体内注射NK-5962, RCS大鼠视网膜外核层的凋亡细胞数量无论在光照或黑暗条件下均有所减少。


在本研究中，我们回顾了NK-5962神经保护作用的体内外证据，并设计了药代动力学实验。


体外IC50为1.7 μM，可以解释玻璃体腔注射对视网膜神经元凋亡的保护作用的体内EC50为3 μM。


NK-5962在大鼠眼内的药代动力学研究表明，静脉给药而非口服能使NK-5962在大鼠眼内有效富集。


NK-5962将成为延迟视网膜营养不良恶化的候选药物，如色素性视网膜炎。
