一、试剂盒简介

引物独立，带DNase的一步逆转录试剂盒为一款基因组DNA去除与逆转录反应在一步进行的新一代快速逆转录试剂盒，与EZBioscience 4xEZscript Reverse Transcription Mix II(with gDNA Remover)(Cat.No.: EZB-RT2G)相比，不需要单独进行去除基因组DNA的反应，操作更简便，并可有效降低复杂加样过程造成的样品污染与RNA降解的风险。试剂盒中主要包括5管试剂:Enzyme Mix中含有DNase、RNaseInhibitor和逆转录酶;5xEZscript All-in-one RT Buffer中含有buffer、dNTPs等;NC Buffer;Oligo dT18(20x)和Random Hexamer(20x)。Oligo dT18和Random Hexamer这两种引物独立包装，便于根据需要添加目标片段特异性引物。

引物独立，带DNase的一步逆转录试剂盒采用的新型M-MLV突变体逆转录酶具有很强的抗干扰能力及扩增能力，同时又采用了新优化的反应体系，进一步提高了逆转录效率，使用本试剂盒逆转录15分钟得到的扩增产物量可以达到AMV逆转录酶大约1小时的产物量(Ct值相同)。后续逆转录产物建议用于PCR、qPCR和基因克隆。

一、试剂盒简介

概述

Monarch 总 RNA 小量提取试剂盒是一款提取并纯化总 RNA 的全能解决方案试剂盒，能做到样本保存、细胞裂解、gDNA 去除，适用于各种生物样本，如培养细胞、血液和哺乳动物组织，也适用于难裂解样本，如细菌、酵母和植物，只要增加一步来提高裂解效率。该试剂盒还可以用于纯化、酶学反应后的样本或者 TRIzol® 提取的样本，提取纯化后的 RNA 具有极高质量：A260/280 和 A260/230 比值均 ≥1.8、高 RIN 值和无 gDNA 残留；并且 RNA 片段包含完整的 miRNAs 和完整的 rRNAs。另外，改变结合条件可以选择性地筛选小于 200 nt 的 RNA，包含miRNA、5S rRNA 和 tRNA。提取纯化得到的 RNA 可用于 RT-qPCR、
cDNA 合成、RNA-seq、Northern blot 分析等下游实验。

概述

DNase I（无 RNase）是一种非特异性核酸内切酶，切割 DNA 产生具有 3´ 羟基和 5´ 磷酸末端的二核苷酸、三核苷酸和寡核苷酸产物。DNase I 可以作用于单链 DNA、双链 DNA、染色质和 RNA:DNA 杂交链。 

来源

重组 E. coli 菌株，含有牛胰腺 DNase I 的 MBP 融合克隆。 

反应条件

1X DNase I 反应缓冲液
[10 mM Tris-HCl (pH 7.6 @ 25℃) ，2.5 mM MgCl2，0.5 mM CaCl2]，37℃ 温育。
热失活：75℃ 10 分钟。 

质保声明

无 RNase 污染。 

单位定义

1 单位指 50 μl 反应体系中，37℃ 条件下，10 分钟完全分解 1 μg pBR322 DNA 所需要的酶量。
完全降解是指绝大多数的 DNA 片段降解成为四核苷酸或更短的核苷酸。 

浓度

2,000 units/ml。 

注意事项

为避免酶失活过程中 RNA 被降解，应添加终浓度为 5 mM 的 EDTA。

产品描述

 DNase I-XT（耐盐）是基于 DNase I 改造的耐盐型 DNA 核酸内切酶，可非特异切割 DNA，产生具有 5′-磷酸和 3′-羟基末端的二核苷酸、三核苷酸和寡核苷酸产物 (1,2)。DNase I-XT（耐盐）作用于单/双链 DNA、染色体，以及 RNA:DNA 杂交链上的 DNA。当盐浓度>50 mM 时，不耐盐型 DNase I（无 RNase）（NEB #M0303）的活性会受到抑制，但 DNase I-XT（耐盐）（NEB #M0570）在 50-100 mM 盐浓度范围活性最佳，盐浓度达到 200 mM 时，可维持 65% 活性，300 mM 时，仍有~40% 活性。

在等摩尔的 DNase I (NEB #M0303) 和 DNase I-XT（耐盐）(NEB #M0570) DNase 活性比较中，DNase I-XT（耐盐）展示出更高的耐盐特性。检测方法为：在 1X DNase I 反应缓冲液中，消化标记有荧光和淬灭基团的发夹 dsDNA 底物（35 nt），荧光强度反映出不同盐浓度条件下的酶活（如图所示）。结果显示：DNase I 活性随着盐浓度的增加而下降，而耐盐的 DNase I-XT 在高达 300 mM 盐浓度中仍保持活性。

DNase I-XT（耐盐） 货 号 #M0570L

20 μl 体外转录产物用以下三种方法检测去除残留 DNA 的效果：1) 无 DNase I；2) 2 U TURBO® DNase 或 3) 2 U DNase I-XT（耐盐），37°C 条件下孵育 15 分钟。分别使用 Monarch RNA 纯化试剂盒（500 µg）（NEB #T2050）纯化，以无核酸酶水（50 µl）洗脱。使用 Luna 通用探针法 qPCR 预混液（NEB #M3004）通过 qPCR 检测 DNA 残留水平。将每个样品的平均 Cq 值与标准曲线（灰色）进行比较，以评估可 PCR-扩增 DNA 的百分比。TURBO DNase 和 DNase I-XT（耐盐）的使用都不需要对 IVT 反应产物进行稀释，但是，DNase I-XT（耐盐）对 IVT 反应中的 DNA 模板去除更彻底，难以通过 qPCR 检测到。

RNA 粗提样本用以下四种方法检测去除残留 DNA 的效果：1) 无 DNase I；2) 在 DNase I-XT 反应缓冲液中加入 DNase I-XT（耐盐）(2 U, NEB #M0570)；3) 在 DNase I 反应缓冲液中加入 DNase I (2 U, NEB #M0303)；4) 在 TURBO DNase 反应缓冲液中加入 TURBO DNase (2 U, ThermoFisher) ，37°C 条件下孵育 15 分钟。使用 Luna 通用一步法 RT-qPCR 试剂盒 (NEB #E3005) 和人类或小鼠 Actin 外显子引物，通过（+/- RT）qPCR 检测对残留的基因组 DNA (gDNA) 进行定量。以 qPCR（无反转录，仅扩增DNA）与 RT-qPCR（有反转录，同时扩增 DNA 和 RNA）平均 Cq 值的差值评估 DNase 去除 DNA 的效果。结果显示：对于 RNA 粗提产物中的残留 gDNA，DNase I-XT（耐盐）比 TURBO DNase 去除效果更彻底。
