Gibco™ Ham's F-12 Nutrient Mix

Using F-12:

Compared to other basal media, F-12 contains a wider variety of components, including zinc, putrescine, hypoxanthine, and thymidine. F-12 contains no proteins or growth factors. Therefore, F-12 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). F-12 uses a sodium bicarbonate buffer system (1.176 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.


cGMP manufacturing and quality system:

For supply chain continuity, we manufacture F-12 at two separate facilities; one located in Grand Island, NY and the other in Scotland, UK. Both sites are compliant with cGMP-manufacturing requirements, are certified to ISO 13485, and are registered with the FDA as medical device manufacturers.

cGMP Manufacturing and Quality System:

Gibco™ F-12 is manufactured at a cGMP compliant facility located in Grand Island, New York. The facility is registered with the FDA as a medical device manufacturer and is certified to ISO 13485 standards.


Gibco™ F-12 contains no proteins or growth factors. Therefore, F-12 requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). F-12 uses a sodium bicarbonate buffer system (1.176 g/L) and therefore requires a 5-10% CO2 environment to maintain physiological pH. Powder forms of Gibco™ cell culture medium require sodium bicarbonate supplementation, pH adjustment, and filtration at the time of preparation.

General description:Media are supplemented with a defined mixture of nutrients, growth factors, and hormones instead of serum. A 1:1 mixture of Dulbecco′s Modified Eagle′s medium (DME) and Ham′s F-12 nutrient mixture is commonly used for these defined studies. To compensate for the loss of buffering capacity caused by eliminating serum, a final concentration of 15 mM HEPES buffer can be added to the formulations. It allows the investigator to optimize the buffering system.

Application:Dulbecco′s Modified Eagle′s Medium/Nutrient Mixture F-12 Ham has been used:

to maintain human lung and colorectal cancer cell lines for the biological evaluation of the secondary metabolites

to culture human kidney-2 (HK-2) cells and as a component of the assay medium for lysophosphatidylcholine (LPC) treatment

to culture mouse squamous cell carcinoma (SCCVII) tumor cells

General description:Pyridoxal, which is an aldehyde, has been replaced with pyridoxine. This modification has been shown to improve the stability of the medium. A measured mixture of chemically defined ingredients makes up a basal medium, which is a type of complex media. DMEM is a medium supplemented with serum and sometimes with tryptose phosphate broth (TPB). DMEM/F-12 is a completely defined synthetic medium, which is protein-free.

Application:

DMEM:F12 is a 50:50 mixture of DMEM and Ham′s F12 media that has proven to be useful in a wide range of cell culture applications, especially when supplemented with fetal bovine serum (FBS). Ham′s Nutrient Mixture F12 was originally developed for the serum-free clonal growth of Chinese Hamster Ovary (CHO) cells, lung cells, and mouse L cells. It is frequently used with dialyzed serum, hormones, selenium, and other supplements for serum-free cultures. It is the medium of choice for supporting the growth of cells of rodent origin, particularly rabbit and rat, and has been proven to be an excellent cloning medium for myeloma and hybridoma cells.

Suitable for growth of wide range of cell types in low serum conditions.

Dulbecco′s Modified Eagle′s Medium/Nutrient Mixture F-12 Ham is used in cell culture & proliferation assays. It is also used in bone marrow-derived macrophages culture.

Dulbecco′s Modified Eagle′s medium (DMEM), a regularly used cell culture medium, is a source of energy-rich media that includes glucose, vitamins, and amino acids. A 1:1 mixture of Dulbecco′s Modified Eagle′s medium (DME) and Ham′s F-12 nutrient mixture is commonly used in cell culture research. To compensate for the loss of buffering capacity caused by removing serum, 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) buffer may be added in the formulations at a final concentration of 15 mM.
