产品说明：多发性骨髓瘤培养基（Human Multiple Myeloma Cell Medium）是一种为促进人源多发性骨髓瘤原代细胞体外生长而设计的*培养基。它是一种无菌的液体混合系统，含有必需和非必需的氨基酸、维生素、有机和无机化合物、生长因子、微量矿物质等。培养基是基于碳酸氢盐的缓冲体系，在37℃、5%CO2的培养箱中平衡时，pH值为7.4。该培养基的配方可以提供一个理想平衡的体外培养环境，选择性地促进人源多发性骨髓瘤原代细胞的生长。

使用说明：
◉样品分离
（1）使用移液器将抗凝管中的外周血或骨髓样本混匀，转移至15mL无菌离心管中，1500rpm离心5min。
（2）弃血浆，向血细胞沉淀中加入3~5倍1×PBS稀释至9mL，充分混匀。
（3）另取15mL离心管，加入6mL人外周血淋巴细胞分离液，倾斜缓慢地加入上述稀释的血细胞沉淀，与分离液明显分层，设置转速400g、升速2、降速0、室温，离心30min。
（4）离心后，离心管中由上至下细胞分四层（PBS层、环状乳白色白细胞层、分离液层、红细胞层），移液器环形吸取白细胞层至预先加入5mL1×PBS的15mL离心管中，轻轻混匀洗涤细胞，1500rpm室温离心3min。
（5）弃上清，加入6mL红细胞裂解液重悬细胞沉淀，4℃裂解15~20min，中间颠倒混匀一次，1500rpm室温离心3min。
（6）弃上清，加入1~5mL*培养基重悬细胞沉淀，活细胞计数后（细胞活率至少>50%），并参考表1，接种活细胞于培养瓶/板，置37℃、5%二氧化碳培养箱中培养。
◉细胞传代
（1）镜下观察细胞形态，待悬浮细胞长满至80%～90%时即可传代。
（2）将细胞悬液转移至15mL离心管，1500rpm室温离心3min。
（3）弃上清，以1~2mL培养基重悬细胞，活细胞计数，并参考表1，接种活细胞于培养瓶/板，置37℃、5%二氧化碳培养箱中培养。
注意事项：
1.本产品在使用前需平衡至室温。
2.原代细胞培养期间尽量避免频繁摇晃，显微镜观察原代细胞状态尽量避免摇晃细胞培养器皿。
3.使用本品恶性浆细胞比例越高（至少>40%），培养成功率越高。4.使用产品时应注意无菌操作，避免污染。
5.本产品中含有血清和原代细胞专用抗生素，如无特别需要不用额外再添加血清和双抗，可直接使用。
6.为保持本产品的理想使用效果，不宜将其过夜放置于室温或较高的温度环境中。7.请于保质期内使用本产品，超出保质期，必须放弃使用。
8.本产品仅用于科研用途，不能用于体外诊断。
