Description:

DBCO (Dibenzocyclooctyne) is a cycloalkyne that can reacts with azides via strain-promoted 1,3-dipolar cycloaddition in aqueous solution, a bioorthogonal reaction also termed Cu-free click reaction. This reaction has excellent selectivity and biocompatibility, such that the complimentary reagents can form covalent bonds with richly functionalized biological systems. The Cu-free click reaction has been a powerful tool in catalyst-free bioconjugation. DBCO reagents posses fast kinetcs and stability in aqueous buffer and they can be used to label azide-modified biomolecules with high specificity and reactivity.

Copper-free Click Reaction Procedures:

Prepare the azide-containing sample in reaction buffer. Add DBCO conjugate to azide containing sample. Recommendation: Add 1 mole equivalent of limiting reagent to 1.5-3.0 mole equivalents of highest abundance reagent. Incubate the reaction at room temperature for 2-4 hours or 2-12 hours at 4 degree celcius. The reaction is now ready for purification.

Proteinase K is a highly reactive nonspecific serine protease that is capable of inactivating RNases and DNases and is used in the isolation or preparation of high molecular weight nucleic acids. Proteinase K is also useful for helping to characterize enzymes, due to its cleavage specificity. Proteinase K is stable in a wide variety of detergents and buffer salts and at various temperatures and pH. The isoelectric point of proteinase K is 8.9. Nanocs provides a number of fluorescent labeled Proteinase K products that span from UV region to near IR. These fluorescent Proteinase K conjugates can be used for enzyme studies, bioassay development and many other biomedical research. Nanocs Cy5 labeled Proteinase K is a near IR fluorescent Proteinase K conjugate that has excitation/emission wavelength at 650/670 nm. This Cy5 Proteinase K can be easily detected by fluorescent instruments in Cy5 channel. All our fluorescent Proteinase K conjugates were purified by size chromatography to remove non-conjugated molecules and ensure adequate applications both in-vitro and in-vivo.
